Evommune, Inc. (NYSE: EVMN ) announced on September 8 that EVO756 missed its primary endpoint in a randomized, double-blind, placebo-controlled Phase 2b trial involving 121 adults with moderate-to-severe atopic dermatitis. The primary measure was the percentage change from baseline in Eczema Area and Severity Index, or EASI, at week 12. No tested dose met the primary or secondary endpoints, and development in that indication will stop.

Evommune, Inc. (NYSE:EVMN) had already discontinued EVO756 for chronic spontaneous urticaria following a June trial failure. Development continues in migraine prevention. The setbacks place greater weight on EVO301, which has positive Phase 2a eczema data and a Phase 2b study planned for mid-2027.

Evommune, Inc. (NYSE:EVMN) has clinical evidence supporting its next eczema program. In February, the company reported that EVO301 met the primary endpoint in a randomized, double-blind, placebo-controlled Phase 2a trial involving 70 adults. Participants received two intravenous doses or placebo over a 12-week study.

Evommune, Inc. (NYSE:EVMN) reported a 55% EASI reduction with EVO301 versus 22% with placebo at week 12, a difference of 33 percentage points. The study met its prespecified Bayesian success criterion, and a separate conventional analysis showed statistical significance at p<0.01. No treatment-related serious or severe adverse events were reported.

EVO301 also gives Evommune, Inc. (NYSE:EVMN) a distinct biological approach. The injectable fusion protein targets interleukin-18, an inflammatory signaling protein, while EVO756 blocks the MRGPRX2 receptor. That distinction supports assessing EVO301 on its own evidence.

The balance sheet provides time for further testing. Evommune, Inc. (NYSE:EVMN) reported $288.0 million in cash, cash equivalents, and investments as of June 30. In the September update, management expected resources to support operations into 2029. Ending unsuccessful eczema development could preserve funding for more promising work.

Two failed indications reduce the number of credible development opportunities for EVO756. Evommune, Inc. (NYSE:EVMN) now needs to demonstrate why continued migraine investment has a better prospect of success. The ongoing Phase 2b trial targets approximately 330 participants, with results expected in 2027. Its outcome remains uncertain.

EVO301 also faces a substantial development step. The Phase 2a study tested intravenous dosing, while Evommune, Inc. (NYSE:EVMN) plans to use subcutaneous regimens in Phase 2b. Manufacturing scale-up and toxicology work precede the planned mid-2027 start. Dose selection, treatment durability and safety across a larger population remain important tests.
